A new assay for quantitative detection of hepatitis A virus

Hepatitis A virus (HAV) is mainly transmitted via contaminated food or water through person-to-person contact. Here, we describe development and evaluation of a reverse transcription droplet digital PCR (RT-ddPCR) real-time (RT-qPCR) assay for detection HAV in clinical specimens. The was evaluated by assessing limit detection, precision, matrix effects, sensitivity quantitative agreement. 95 % (LOD95 %) 10 higher RT-ddPCR than RT-qPCR. Bayesian model used to estimate precision on different target concentrations. From this, found that had somewhat greater RT-qPCR within runs markedly between runs. By analysing serum from naturally infected persons sample, the two methods agreed well quantification comparable sensitivities. Tests with artificially samples revealed neither nor severely inhibited presence oysters, raspberries, blueberries leafy-green vegetables. For this assay, conclude should be considered if rapid, qualitative main interest precise interest. high allows small changes viral concentration over time, which has direct implications both control studies.
